Similar to the GBA original, the storyline revolves around Donkey Kong's theft of all the Mini-Mario toys, requiring players to guide Mario through a series of platforming puzzles to retrieve them.

The remake incorporates numerous new features to captivate existing fans and help new ones grasp the game's unique gameplay. Among these is a brand-new local co-op mode for Mario vs. Donkey Kong, enabling one player to control Mario while the other controls Toad. The latest trailer provides a glimpse of these new features and the additional content designed to enhance the original game.

This Mario vs. Donkey Kong trailer confirms the inclusion of 130 levels in the game, an increase from the original 90 levels and 12 expert stages. The original game featured these levels across six distinct world themes, including two boss stage-only levels against DK. The expanded 130 levels comprise two new world themes in the remake, namely an icy cave named Slippery Summit and a circus fairground called Merry Mini-Land. These worlds introduce their own unique elements, such as slippery ground, icicle spikes, and directional flower gusts. Additionally, expert levels, which can be unlocked, aim to challenge players' skills after achieving a certain number of Perfect rating stars, with 16 unlockable expert levels featured in the remake.
